UniMab EXE Protein A Affinity Chromatography Resin

To address the challenges of traditional agarose-based Protein A affinity resins in scale-up production, NanoMicro leverages its technical expertise in "rigid" media development and adopts a "rigid-flexible combined" design philosophy to launch UniMab® EXE, a next-generation high-performance, alkali-tolerant recombinant rProtein A affinity chromatography resin.

The UniMab® EXE matrix features a core-shell structure with a rigid, monodisperse porous polymethacrylate (PMMA) microsphere core, providing superior mechanical strength and pressure resistance for a more stable and higher column bed. A flexible dextran coating is applied to the surface and internal pores of the PMMA microspheres, and the multi-hydroxyl structure ensures excellent hydrophilicity, reducing non-specific binding. The optimized Protein A ligand density enables higher binding capacity and increased antibody batch throughput. Through this unique and innovative design, UniMab® EXE significantly improves antibody production efficiency and reduces costs, meeting the demands of laboratory-scale preparation, pilot-scale, and industrial-scale production (as illustrated).

UniMab® EXE maintains high dynamic binding capacity for conventional monoclonal antibodies and Fc-containing recombinant proteins at high flow rates. It also efficiently captures next-generation bispecific (multi-specific) antibodies and antibody-drug conjugates, greatly improving production efficiency and reducing antibody production costs.

Compared to similar products on the market, it offers the following unique advantages:

  • Tolerates 0.1-0.5 M NaOH cleaning, providing long service life and low production costs; binding capacity loss is less than 10% after 24 hours of exposure to 0.5 M NaOH;

  • High binding capacity: average dynamic binding capacity for mAb is approximately 55-65 mg·mL⁻¹ (4 min residence time); DBC for bispecific antibodies is >40 mg·mL⁻¹ (5 min residence time);

  • Low host cell protein (HCP) residuals: typically below 1000 ppm;

  • High recovery: greater than 90%;

  • High purity: antibody purity in the affinity eluate exceeds 99%;

  • Mild elution conditions, making it an ideal choice for antibody purification platform processes.
