NMOligo Series Solid Phase Synthesis for Oligonucleotides

NMOligo is a high-performance solid-phase synthesis support developed by NanoMicro based on years of PS-DVB polymer synthesis technology and surface modification methods. Utilizing NanoMicro's proprietary pore-forming technology, this support features uniform pore size and particle size distribution as well as an optimized cross-linked polystyrene structure, enabling high-purity, high-yield, and high-loading oligonucleotide synthesis while reducing solid-phase synthesis costs.

Product Description

Introduction to Nucleic Acid Solid-phase Synthesis Supports

Nucleic acid solid-phase synthesis supports are critical materials in the chemical synthesis of oligonucleotides (oligonucleotide drugs). They are used to immobilize the initial nucleotide, providing a stable foundation for subsequent complex coupling reactions, ensuring that each nucleotide is accurately connected to progressively build oligonucleotides with specific sequences and functions. Currently, the phosphoramidite solid-phase synthesis method is predominantly used for oligonucleotide synthesis. Each complete synthesis cycle involves four steps and adds one nucleotide residue to the growing oligonucleotide chain attached to the solid support.

Currently, there are two mainstream support matrices on the market:

One is controlled pore glass (CPG) with inorganic matrix. In research settings, CPG is widely used, especially showing advantages in synthesizing longer nucleotide chains. However, CPG has loading capacity limitations, typically only 50-150 μmol/g, and is fragile, which poses constraints in scale-up synthesis and makes it difficult to meet large-scale production demands.

The other is polystyrene microspheres (PS-DVB) with organic matrix. The key feature of PS-DVB is its high loading capacity, generally reaching 350 μmol/g or above. Additionally, as an organic matrix, it offers good toughness, is not easily broken, and is widely used in large-scale synthesis.

DNA Synthesis on NMOligo-Unylinker350

The performance of a solid support is the starting line of the synthesis process and has a major impact on the yield and purity of the final product. In a side-by-side comparison under identical conditions, a 20-base oligonucleotide and a short-chain DNA (Gapmer) were synthesized on a support from a leading international brand and on the NanoMicro support. Comparison of the crude purity and crude yield of the oligonucleotides shows that the NanoMicro support matches the performance of the leading international brand.
