Introduction
Caspofungin is an echinocandin antibiotic, a novel lipopeptide antifungal drug semi-synthesized from Pneumocandins B0, the fermentation product of the Glarea Lozoyensis strain. Caspofungin exerts its antifungal activity by inhibiting the synthesis of β(1,3)-D-glucan, an essential component of the cell wall of many filamentous fungi and yeasts.

Challenges in Caspofungin Purification
In recent years, the market for echinocandin antifungal drugs such as anidulafungin, caspofungin, and micafungin has grown rapidly. However, the requirements for caspofungin, a fermentation-derived semi-synthetic product, are extremely stringent. Systematic studies on strain cultivation and fermentation processes are essential, along with the exploration of synthesis routes, conditions, and subsequent purification processes. Given the fragile nature of the final product and the difficulty in controlling impurities during the process, significant challenges are added to downstream purification process development and control.
Purification Results
NanoMicro has developed a dedicated chromatography purification process for caspofungin, offering high loading capacity, excellent product purity, and superior control of individual impurities. The process employs isocratic elution, ensuring simple process control and low equipment requirements.
